Định danh vi sinh vật bằng công nghệ khối phổ protein MALDI TOF MS

Phân lập và định danh vi sinh vật là phương pháp vô cùng quan trọng trong chẩn đoán và thử nghiệm vi sinh vật nhằm xác định nguyên nhân gây bệnh. Tuy nhiên, hiện nay phương pháp truyền thống như định danh bằng phản ứng sinh hóa cho kết quả chẩn đoán sau 3 – 5 ngày, độ chính xác còn một số hạn chế, ảnh hưởng đến việc xác định kịp thời nguyên nhân gây bệnh do vi sinh vật. Ngoài ra, phương pháp truyền thống cần giám định kết quả bằng kỹ thuật PCR.

Để khắc phục những nhược điểm từ phương pháp truyền thống trên và đáp ứng nhu cầu nghiên cứu, phục vụ chẩn đoán dịch bệnh, Phòng thí nghiệm Trọng điểm Công nghệ sinh học Thú y, khoa Thú Y –  Học viện Nông nghiệp Việt Nam thông qua nguồn kinh phí dự án “Đầu tư cơ sở hạ tầng, trang thiết bị phục vụ kiểm soát dịch bệnh lây truyền giữa động vật, người và nâng cấp khu nuôi động vật thí nghiệm đạt chuẩn an toàn sinh học cấp III” của Bộ Nông nghiệp và Phát triển Nông thôn đã đầu tư trang thiết bị và ứng dụng công nghệ hiện đại MALDI TOF MS (Matrix assisted laser desorption ionization – time of flight) để mở rộng năng lực nghiên cứu trong việc định danh nhanh, chính xác vi sinh vật, đặc biệt các vi khuẩn gây bệnh trên vật nuôi (bao gồm cả các vi khuẩn gây bệnh truyền nhiễm trên động vật và người và bệnh mới nổi). MALDI TOF MS sử dụng công nghệ khối phổ protein định danh vi sinh vật, so sánh sự tương đồng của phổ protein từ mẫu vi sinh vật mục tiêu với cơ sở dữ liệu của hơn 8000 chủng vi sinh vật khác nhau. Phân tích MALDI TOF MS cho phép định danh chính xác loài vi sinh vật, bao gồm vi khuẩn Gram âm, Gram dương, vi khuẩn kỵ khí, vi khuẩn hiếu khí, nấm men, nấm mốc, Mycobacteria. Hệ thống máy yêu cầu cán bộ nghiên cứu chuyên nghiệp, có trình độ cao, thành thạo tay nghề, cho phép định danh vi sinh vật trực tiếp từ môi trường thạch hoặc mẫu bệnh phẩm trong thời gian 30 phút với 96 mẫu vi sinh vật. Công nghệ hiện đại khối phổ protein cho kết quả chẩn đoán nhanh vi sinh vật, độ chính xác cao, quản lý dữ liệu, cập nhật kết quả mới thường xuyên từ ngân hàng vi sinh vật thế giới, kết quả có giá trị khoa học cao.

Ngoài việc phục vụ công tác nghiên cứu chuyên sâu tại phòng thí nghiệm, các đơn vị nghiên cứu, doanh nghiệp, cán bộ kỹ thuật, người chăn nuôi khi có nhu cầu phân lập và định danh chính xác các vi sinh vật, Phòng thí nghiệm Trọng điểm Công nghệ sinh học Thú y, Khoa Thú Y –  Học viện Nông nghiệp Việt Nam còn đáp ứng yêu cầu thực hiện việc thử nghiệm, trả kết quả phân lập và định danh nhanh vi khuẩn trong 24 – 36 giờ từ đó kịp thời đánh giá kháng sinh đồ để đưa ra phác đồ điều trị hiệu quả đem lại ý nghĩa quan trọng trong công tác chẩn đoán nhanh và phòng, chống dịch bệnh góp phần nâng cao năng suất và phát triển chăn nuôi bền vững.
